    • Effects of resveratrol on multidrug resistance in human retinoblastoma cells

      Objective To observe the effect of resveratrol on multidrug resistance (MDR) in human retinoblastoma cells treated. Methods RB cells in logarithmic growth phase were divided into experimental group and control group. RB cells in experimental group were cultured with different concentrations of resveratrol (6.25, 12.50, 25.00, 50.00, 100.00 mu;mol/L) for 24 and 48 hours. The proliferation (absorbance value) was assayed using methyl thiazolyl tetrazolium (MTT). RB cells were cultured with 50.00 mu;mol/L resveratrol for 48 hours. The expressions of MDR-1, cyclooxygenase-2 (COX-2)、multidrug resistance-associated protein-1 (MRP-1), glutathione-S-transferases-pi; (GST-pi;) were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot. The RB cells of the control group were cultured with 0.5% dimethyl sulfoxide. Results Compared with the control group, the absorbance value decreased in experimental groups (6.25, 12.50, 25.00, 50.00 mu;mol/L) in a dose dependent manner (F=4.782,P<0.05). The difference of absorbance value between 50.00 and 100.00 mu;mol/L experimental groups was not significant (F=6.351,P>0.05). Compared with the control group, the mRNA (t=9.170, 5.758, 4.152, 4.638) and protein (t=3.848, 5.955, 4.541, 3.514) expression levels of MDR-1, MRP1, COX-2, and GST-pi; decreased in the experimental group (P<0.05). Conclusion Resveratrol can down-regulate the expression of MDR in RB cells.

    • Expression of sperm protein 32/OY-TES-1 in retinoblastoma

        Objective To observe the expression of sperm protein (SP) 32/OY-TES-1 in retinoblastoma (RB).Methods Thirty paraffin specimens of pathologically confirmed RB eyeballs were investigated in this study. The SP32/OY-TES-1 mRNA expression of 15 RB tissues, 12 non-tumor retinal tissues and 22 normal eye tissues were detected by reverse transcription polymerase chain reaction (RT-PCR). The SP32/OY-TES-1 protein expression of 30 RB tissues and 24 normal retinal tissues were examined by immunohistochemistry. The relationship between SP32/OY-TES-1 mRNA, protein expression and age, gender, tumor size, tumor differentiation, clinical stage of RB children were analyzed. Results The expression rate of SP32/OY-TES-1 mRNA in RB tissues, non-tumor retinal tissues and normal eye tissues were 86.7%, 16.7% and 36.4% respectively. Compared the expression rate of SP32/OY-TES-1 mRNA in different gender (chi;2=0.744),age(chi;2=1.178),clinical stage(chi;2=1.188),tumor size (chi;2=0.216),tumor differentiation(chi;2=1.885),the differences were not statistically significant (P>0.05). The expression rate of SP32/OY-TES-1 protein in RB tissues was 73.3% and no expression in normal retinal tissues. Compared the expression rate of SP32/OY-TE-1 protein in different gender (chi;2=1.000),agewith le;two years and >two years(chi;2=0.403),tumor size (chi;2=2.274),tumor differentiation(chi;2=0.138), the differences were not statistically significant (P>0.05); but in clinical stage (chi;2=6.193),with or without optic nerve infiltration (chi;2=4.535), the differences were statistically significant(P<0.05). Conclusions SP32/OY-TES-1 is highly expressed in RB. The expression rate of SP32/OY-TES-1 is related to optic nerve infiltration and clinical stage of RB.

    • Diagnosis of retinoblastoma by ultrasonography

        Objective To explore the characteristics and diagnostic values of ultrasound examination of retinoblastoma (RB).Methods The ultrasound and CT features of 210 eyes (162 patients) with pathologically confirmed RB were analyzed retrospectively. Results The ultrasonography image of those RB eyes were all characterized by substantial masses in the posterior segment of the eyeball, shown as spherical, hemispherical and irregular in shape, and even filled the entire eyeball. Calcification within the mass was observed in 197 eyes of 149 patients (92.0%), but not observed in 13 eyes of 13 patients (8.0%).Colorful blood flow signals extended from the central retinal vessels could be seen inside the masses of all patients. Ultrasound diagnosis was consistent with the pathological diagnosis in 92.0% RB cases. CT examination revealed calcified speckles or plaques in 167 eyes from 145 patients (89.5%), consistent with the pathological diagnosis of RB.Conclusions Ultrasonography can show the tumorprime;s shape, size, internal features and the range of orbital involvement. It is a valuable clinical tool in the diagnosis of RB.

    • THE RELATION BETWEEN RETINOBLASTOMA GENE EXPRESSION AND BIOLOGICAL CHARACTERISTICS OF GALLBLADDER CARCINOMA

      Objective To study the relation between retinoblastoma (Rb) gene expression and biological characteristics of gallbladder carcinoma. Methods The expression of Rb protein in tissues of 41 cases of gallbladder carcinoma, 7 cases of gallbladder papilloma, and 14 cases of cholecystitis were detected by immunohistochemical staining of SP with polyclonal antibody. Results The rate of positive staining was 58.7% in gallbladder carcinoma which was significantly lower than that in cholecystitis and gallbladder papilloma (100%). There was a significant difference of Rb protein expression among the low, moderate and high differentiations and so was between S5, S4, S1, S2 and S3 stage (P<0.05). Conclusion It sugests that expression or non-expression of Rb gene is closely related to the malignant potential invasiveness and prognosis of gallbladder carcinoma.

    • Relationship of differentiated degree with spread or survival prognosis in retin oblastoma

      Objective To probe the relationship of differentiation degree with spread or survival prognosis in retinoblastoma (RB). Methods Clinical data, follow up status and eyeball specimens in 156 RB cases were investigated retrospectively. The tumors were divided into differentiated and undifferentiated groups. Conditions of the tumor invasion of ocular or surrounding tissues were reviewed. The fatality rate was obtained from the follow-up materials of 82 cases of RB. The fatality rate and the invasion rate between the two types were compared statistically by Chi-square test. In addition, the relation between the tumor invasion and death ,and the average survival time for dead people after surgery were explored. Results Local invasion of tumor cell was found in 8 eyes among 17 eyes with differentiated RB (47.06%),and in 66 eyes among 139 eyes with undifferentiated RB (47.48%).There was no significant difference with regards to the local invasion between the two types ( The fatality rate of cases of differentiated RB was 27.27%,and 22.54% in undifferent iated RB, and there was no statistical difference between the two types .The fat ality rate for patients with orbital and scleral extension was 100%, optic nerve invasion (grade Ⅳ) was 62.50%,and uveal invasion was 22.22%.The survival time for the dead victims were from 5 months to 41 months and averaged to 21.92 months. Conclusion There was no significant differ ence both in survival prognosis and local invasion between the two types. The survival prognosis of metastatic RB was dependent on the degree of spread and the efforts of treatment and regardless of the types of differentiation of RB cells. (Chin J Ocul Fundus Dis, 2001,17:18-20)

    • ESTABLISHMENT OF A VINCRISTINE-RESISTANT HUMAN RETINOBLASTOMA CELL LINE

      PURPOSE:Studying the multidrug resistance(MDR) phenotype occurring in retinoblastoma and its mechanism. METHODS:Using the procedure of stepwise increase in drug concentrations to obtain a retinoblastoma subline which resistant to 600ng/ml vincristine (HXO-RB/VCR). Characteristics of this drug-resistant cell line were investigated by cell counting,drugcontents determinatin,drug sensitivity evaluation and radiation sensitivity test. RESULTS:This cell line was cross-resistant to VDS,MMC VP16,ADM ,DDP,CBP,but not resistant to BCNU and 5-Fu. It was proved to be collaterally sensitive to MTX,and the response to 60Co gamma;-ray was modified slightly in HXO-RB/VCR cell line. Intracellular levels of VCR was much higher in HXO-RB44 cells than in the resistant subline. Those cross-resistances can be reversed by verapamil partly. CONCLUSIONS:MDR and radiation resistance of retinoblastoma can be induced by exposing to VCR and reversed by verapamil partly. (Chin J Ocul Fundus Dis,1997,13: 6-9)

    • The effects of melatonin on the proliferative activity of retinoblastoma cell line HXO-RB44

      Objective To investigate inhibited effects of melatonin (MLT) on proliferative activity of retinoblastoma cell line HXORB44 and its related mechanism. Methods HXO-RB44 cells were treated by MLT of different concentration (10-10, 10-9, 10-8, 10-7 mmol/L. Cell counting and tetrazolium dyereduction assay (MTT) were used to determine the effect of MLT on the survival and proliferation of HXO-RB44 cells. Apoptotic nuclei were further analyzed by HoechstPI fluorescence staining. Flow cytometry was used to measure the fluorescent intensity of ROS, cell cycle distribution and apoptosis. Results 10 -6 mmol/L (or exceed) of MLT could inhibit the proliferation of HXO-RB44cells in vitro while 10-7 mmol/L (or below) of MLT couldn't. With the increase of MLT concentration from 10-10 mmol/L to 10-7 mmol/L, HXO-RB44 cells gradually increased the expression of ROS. Hoechst staining showed that 4, 8, 12 and 24 hours after the incubation with MLT, the nuclear pyknosis and nuclear fragmentation increased in HXORB44 cells. The extent of apoptosis was proportional to the concentrations of MLT. Flow cytometry revealed that with the increasing of MLT concentration, G0/G1 and G2/M phase cells increased, S phase cells decreased. The apoptotic rate was also increased. Conclusion 10 -6 M of MLT could inhibit the proliferation of HXO-RB44 cells. This effect may relate to the increased ROS expression, cell cycle arrest at G0/G1 phase and apoptosis of HXO-RB44 cells.

    • Clinical features, treatment and prognosis analysis of the asymptomatic patients with retinoblastoma

      ObjectiveTo analyze the clinical features, treatment and prognosis of asymptomatic patients with retinoblastoma.MethodsA retrospective series of case study. Eight asymptomatic patients (11 eyes) with the diagnosis of retinoblastoma by screening enrolled in Department of Ophthalmology of The Eye-ENT Hospital of Fudan University from January 2006 to March 2019 were included. There were 6 males and 3 females ranging from 2 days to 20 months, with a median age of 6 months. Five patients were unilateral retinoblastoma while 3 patients were bilateral. Based on the International Classification of Intraocular Retinoblastoma, 4 eyes were stage A, 3 eyes were stage B and 4 eyes were stage C. One patient had family history. Four patients were evaluated the Rb1 mutation. Routine ophthalmic examinations and ultra-wide field fundus imaging were performed on the 16 parents and 3 siblings of the 8 patients. Systemic intravenous chemotherapy was performed using the Carboplatin, Vincristine, Etoposide protocol, intra-arterial chemotherapy using Carboplatin and Melphalan, and local treatment involved cryotherapy and transpupillary thermotherapy. The mean follow-up time is 47.25 months.ResultsNone of the 8 children had any ocular symptoms. Six patients received intravenous chemotherapy (5-6 times), 1 patient received intra-arterial chemotherapy (3 times), and 1 patient just received local treatment. Among the 11 eyes, 9 eyes were treated with local cryotherapy and 8 eyes were treated with transpupillary thermotherapy. During the follow-up period, 2 patients had new tumor, and the average time was 6.3 months after the last chemotherapy. At the last follow-up, the tumor disappeared in 11 eyes, remained stable in 11 eyes. The eye protection rate was 100% (8/8) for patients without eyeball excision. The best corrected visual acuity was 0.1 for 3 eyes and 1.0 for 5 eyes. Three eyes were not found. One heterozygous mutation of Rb1 gene [1c.35_69del (p.T12fs)] was identified in 1 patient, and the other 3 patients were not detected. One had bilateral bulbar tuberculosis of the 16 parents, 1 had bilateral RB of the 3 siblings. They were the mother and brother of a child with bilateral RB.ConclusionsFundus screening is helpful for the detection of early RB. The eye protection rate is high and the long-term vision prognosis is good after systemic or topical chemical drugs (IVC, IAC) and ocular topical treatment (cryopreservation and transpupillary thermotherapy).

      Despite its low incidence, retinoblastoma and its rela ted gene (Rb gene) have attracted some of the most brilliant minds in medicine and biology fi elds over the past years. Great advances have been achieved in the tumoregenesis mechanism and clinical management of retinoblastoma recently. However as always , more questions arise from those results. In order to improve retinoblastoma re search in China, we need to strengthen the communication and cooperation with di ffe rent countries, different institutes and disciplines, and utilize the great reso urces of retinoblastoma patients in China.

    • Expression of DNA methyltransferases 1, 3a, and 3b in retinoblastoma

      Objective To observe the expressions of DNA methyltransferases (DNMTs) 1, 3a and 3b in retinoblastoma (RB). Methods Sixty-two RB samples and six normal retinas were studied, including 17 poorly differentiated and 45 well differentiated samples; 16 invasive and 46 non-invasive samples. The expressions of DNMT1, 3a, and 3b, and Ki-67 were detected using immunohistochemical analysis. Brown staining of nuclei was considered to represent the positive stain for DNMT1, 3a and 3b, and ki-67, blue staining as negative. The level of high expression of nuclear staining was, positive cells in DNMT1ge;65%, in DNMT3age;60% and in DNMT3bge;40%. The correlations of DNMT1, 3a and 3b expression in RB samples, and MIB-1 labeling index were analyzed. Results Viewed under the light microscope, negative expressions of DNMT1, 3a and 3b were demonstrated in normal retinas, however, positive expression was observed in RB samples, with 100% in DNMT1, 98% in DNMT3a and 92% in DNMT3b. Comparing well differentiated RB samples with poorly differentiated samples, significant differences were found in high expression of DNMT1 (chi;2=12.57,P<0.05) and DNMT3a (chi;2=10.54,P<0.05); also in the positive cells of DNMT1 (U=179,P<0.05) and DNMT3a (U=198,P<0.05). No significant difference was found comparing high expression (chi;2=1.5,P>0.05) and positive cells (U=307,P>0.05) of DNMT3b. When comparing invasive tumor tissues with non-invasive tumors, significant differences were shown between high expression (chi;2=4.72,P<0.05) and positive cells comparing DNMT1 (U=236,P<0.05). No significant difference was shown in high expression (chi;2=3.53,0.84; P>0.05) in DNMT3a and DNMT3b, or in comparison with positive cells (U=338,257;P>0.05). The expression of DNMTs was positively correlated with the MIB-1 labeling index in RB tissues (R2=0.554,0.376,0.219;P<0.05). Conclusion There are high expressions of DNMT1,3a,and 3b in RB.
