    • Relationships Between Expressions of Somatostatin Receptor Subtypes and Angiogenesis in Colorectal Cancer

      Objective To study the relationships between expressions of somatostatin receptor subtypes(SSTR1-SSTR5) and angiogenesis in colorectal cancer. Methods The expressions of SSTR1-SSTR5, VEGF, and CD34 in the paraffin sections of colorectal cancer tissues from 127 cases were detected by the standard streptavidin-peroxidase (SP) technique. CD34 was used as a marker to account microvessel density (MVD) in colorectal cancer tissues. The relationships between the expressions of SSTR1-SSTR5 and VEGF expression, or MVD were analyzed. Results The positive expression rate of SSTR1, SSTR2, SSTR3, SSTR4, and SSTR5 was 64.6% (82/127), 36.2% (46/127), 18.9% (24/127), 18.9% (24/127), and 38.6% (49/127) in colorectal cancer tissues, meanwhile, the positive expression rate of VEGF was 63.8% (81/127) and MVD was (34.67±16.62)/HP in colorectal cancer tissues. The positive expression rate of VEGF (47.8%, 22/46) and MVD 〔(29.00±15.32)/HP〕 in colorectal cancer tissues with SSTR2 positive expression were significantly lower than those in colorectal cancer tissues with SSTR2 negative expression 〔72.8%, 59/81; (37.90±16.56)/HP〕, Plt;0.05. There were no relationships between SSTR1, SSTR3, SSTR4, and SSTR5 expression and VEGF expression or MVD (Pgt;0.05). Conclusion The positive expression of SSTR2 is related with angiogenesis in colorectal cancer tissues.

    • CULTIVATION AND DIFFERENTIATION OF SPINAL CORDDERIVED STEM CELLS INVITRO IN RATS

      Objective To investigate the method of cultivation and the feature of differentiation of spinal cordderived stem cells in vitro.Methods The neural stemcells from spinal cord of 15 days fetal rats were harvested and cultivated in aserumfree limited medium. The stem cells were induced to differentiate and theresults were identified by cellular immunohistochemistry. Results Lots of stem cells were obtained from the spinal cord of fetal rats and the sphere of stemcells was formed about 10 days. Neural stem cells can give rise to mature neurons and astrocytes.Conclusion Epidermal growth factor/basic fibroblast growth factor serum-free limited medium can promote the proliferation activity ofthe stem cells. Spinal cord-derived stem cells can differentiate into glial cells and neurons.

      Tostudycorrelationbetweenexpressionofp21rasandnm23H1geneandstatusoflymphnodemetastasis(LNM)ingastriccancer.Therateofpositiveexpressionofp21rasandnm23H1werestudiedin80casesbyLSABimmunohistochemicaltechnique.Results:Theresultsshowedthatthepositiveexpressionofp21rasincaseswithLNMwas62.5%,higherthanthatofcaseswithoutLNM(42.5%)(Plt;0.01);thepositiverateofnm23H1incaseswithLNMwas27.5%,lowerthanthatofcaseswithoutLNM(47.5%)(Plt;0.01).Thepositiverateofp21rasincaseswith1-3LNMwaslowerthanthatofcaseswith4-7orgt;8LNM(Plt;0.01);thepositiverateofp21rasincaseswithN1waslowerthanthatofcaseswithN2orN3(Plt;0.01),therewasnopositiveexpressionofp21rasincaseswithN3(7cases).Thepositiverateofnm23H1was44.4%incaseswith1-3LNM,higherthanthatofcaseswith3-7(25.0%)orgt;8LNM(0%)(Plt;0.01),thepositiverateofnm23H1incaseswithN1wasmarkedlyhigherthanthatofcaseswithN2orN3,therewasnopositiveexpressionofnm23H1incaseswithN3(6cases).Conclusions:Theresultssuggestthattheexpressionofp21rasandnm23H1ingastriccancerplayanimportantroleintheinvasiongrowthandmetastasisoftumor,andmayserveasamarkerofpredictingmetastasisandprognosisofgastriccancer.

    • Expressions of Galectin-3 and CD105 Protein in Colorectal Cancer and Their Significances

      ObjectiveTo explore the expressions of galectin-3 protein and CD105 protein in colorectal cancer and the relationship with clinicopathologic features. MethodsThe expressions of galectin-3 protein and CD105 protein 〔microvessel density (MVD)〕 were detected in 60 cases of colorectal cancer tissues, 30 cases of adenoma tissues, and 30 cases of normal mucosa tissues (at least 4 cm far from carcinoma) by MicrowaveEliVisionTM immunohistochemistry, and the relationship with clinicopathologic features was analyzed. ResultsThe expressions of galectin3 protein and MVD in normal mucosa tissues, adenoma tissues, and cancer tissues gradually increased (Plt;0.05). The expression of galectin-3 protein and MVD in colorectal cancer tissues were correlated to TNM stage, invasive depth, and lymph node metastasis (Plt;0.05, Plt;0.01), and the expression of glectin-3 protein was also correlated to differentiated degree (Plt;0.05). The expression of galectin-3 protein in colorectal cancer tissues was positively correlated to MVD (r=0.420, Plt;0.01). ConclusionsThe high expressions of galectin-3 protein and CD105 protein are correlated to the high invasion ability and lymph node metastasis, which may be potential sensitive index to predict the invasion and metastasis of colorectal cancer.

    • THE RELATION BETWEEN RETINOBLASTOMA GENE EXPRESSION AND BIOLOGICAL CHARACTERISTICS OF GALLBLADDER CARCINOMA

      Objective To study the relation between retinoblastoma (Rb) gene expression and biological characteristics of gallbladder carcinoma. Methods The expression of Rb protein in tissues of 41 cases of gallbladder carcinoma, 7 cases of gallbladder papilloma, and 14 cases of cholecystitis were detected by immunohistochemical staining of SP with polyclonal antibody. Results The rate of positive staining was 58.7% in gallbladder carcinoma which was significantly lower than that in cholecystitis and gallbladder papilloma (100%). There was a significant difference of Rb protein expression among the low, moderate and high differentiations and so was between S5, S4, S1, S2 and S3 stage (P<0.05). Conclusion It sugests that expression or non-expression of Rb gene is closely related to the malignant potential invasiveness and prognosis of gallbladder carcinoma.

    • Ultrastructural changes and expression of basicfibroblast growth factor of diabetic retinopathy

      ObjectiveTo investigate relationship between ultrastructural changes and expression of basic fibroblast growth factor of diabetic retinopathy in rats.MethodsDiabetes was induced in rats with a single injection of streptozotocin (STZ) and divided into normal control group and 1- , 3- and 5- month diabetes group. The paraffin slide was observed by in-situ hybridization and immunohistochemistry, and retinal ultrastructure was examined by transmission electron microscopy.ResultsNo change of retinal ultrastructure was found in the control group. Different degrees of ultrastructure lesion were found in 1-month diabetic rats with fragmental increase of thickness of basement membrane, swelling of endothelial cells and obvions fingerlike processes in the capillary cavity, disconcentration of heterochromatin both in endothelium and pericyte, and swelling and degeneration of mitochondrion. The edema of endothelial cells of 3-month diabetic rats was more serious than that of 1month ones, and the capillary cavity was nearly occluded. In 5-month diabetic rats, the basement membrane was unevenly thickened, or obviously split. The positive rate of in-situ hybridization in 3-month diabetic rats was 77.8% while the positive rate of immunohistochemical stain was 55.6%, which increased to 88.9% in 5-month diabetic rats.ConclusionsThe occurrence of the ultrastructural changes in STZ rats with diabetic retinopathy is earlier than that of the expression of bFGF.(Chin J Ocul Fundus Dis, 2003,19:348-351)

    • Significance of p16 and Rb Gene Protein Expression in Primary Gallbladder Carcinoma

      Objective To study the expression of p16 and Rb gene in primary gallbladder carcinoma (PGC). MethodsFiftysix cases of PGC and 26 of gallbladder benign lesions were observed by immunohistochemistry (SP). ResultsThe positive rates of p16 and Rb protein in PGC were 46.3% and 71.4% respectively. No association of Rb expression with histological grade, histological type, invasion, metastasis and prognosis was found. There were close correlation between p16 expression and invasion, metastasis as well as prognosis. The inverse correlation of p16 and Rb gene expression in PGC was confirmed. Conclusion The loss of p16 or Rb gene protein is an important molecular event in PGC,and closely related to the block in the negative regulatory pathway of all cell cycle. The detection of p16 protein may be useful in evaluating biological characteristics and prognosis in PGC.

    • Study the Different Biology Behavior of Typical Breast Medullary Carcinoma and Atypical Medullary Carcinoma

      ObjectiveTo study the cause of different prognosis in typical medullary carcinoma and atypical medullary carcinoma. MethodsThe immunohistochemical staining method was used to evaluate the positivity of Ecadherin,βcatenin, in 30 cases of atypical medullary carcinoma and 18 cases of typical medullary carcinoma and 10 cases of normal breast. ResultsThe positive rate and staining intensity of Ecadherin and βcatenin oncoprotein were significantly higher in typical medullary carcinoma than in atypical medullary carcinoma(Plt;0.01).ConclusionExpression of Ecadherin and βcatenin is one of the causes of similar morphology and different prognosis in medullary breast carcinoma.

    • EXPRESSION AND SIGNIFICANCE OF BASIC FIBROBLAST GROWTH FACTOR IN OVARIAN EPITHELIAL NEOPLASM

      OBJECTIVE To determine the effect of basic fibroblast growth factor (bFGF) on the biological behaviour of ovarian epithelial neoplasm. METHODS Ten cases of normal ovarian tissues and eighty cases of ovarian epithelial tumor tissues were detected by immunohistochemical methods. Mias-2000 Picture Analysis System was used to study the relationship of bFGF expression intensity and microvessel count, FIGO stage, pathological grade and classification of ovarian epithelial neoplasm. RESULTS 1. Expression of bFGF was mainly in cytoplasm and nucleus in several cells of borderline and malignant tumor. 2. The expression intensity of bFGF was closely related to the malignant degree of ovarian epithelial neoplasm. The density of bFGF expression was (3.35 +/- 3.52)% in normal ovarian epithelium, (19.25 +/- 21.73)% in benign tumor, (33.78 +/- 10.86)% in borderline tumor and (48.18 +/- 12.93)% in malignant tumor. The results indicated that bFGF might play an important role in carcinogenesis of ovarian epithelial neoplasm. 3. The expression intensity of bFGF was increased with the FIGO stage of ovarian tumor. 4. The expression intensity of bFGF was increased accompanying with the decrease of differentiation degree in ovian neoplasm. 5. In borderline tumor, expression intensity of bFGF in serous cystadenoma was significantly higher than in mucinous cystadenoma, which indicated bFGF might be an important factor in canceration of ovarian epithelial neoplasm. CONCLUSION bFGF may play important roles in carcinogenesis, development, invasion and metastasis of ovarian epithelial neoplasm.

    • An immunohistochemical study of adhesion molecules in human epiretinal membranes

      Purpose To investigate the expression of intercellular adhesion molecules ICAM-1 and Mac-1,in epiretinal membanes (ERM) of eyes wi th proliferative vitreoretinopathy (PVR). Methods Twenty epiretinal membranes were obtained from eyes undergone vitrectomy for retinal detachment complicated with PVR and observed by immunohistochemical examination. Results Expressions of ICMA-1 and Mac-1 were observed in 18 and 15 membranes respectively.Expression of both adhesion molecules in 12 membranes. Conclusion The findings indicate that adhesion molecules might be involved in the development of PVR. (Chin J Ocul Fundus Dis,2000,16:71-138)
