【Abstract】Objective To investigate the protective effects of epidermal growth factor (EGF) on pancreas of rats with acute pancreatitis(AP). Methods Seventytwo male SpragueDawley rats were randomly divided into 3 groups: Control group, AP group and AP-EGF group. Subcutaneously injection of EGF (0.1 μg/g) were given to animals in the AP-EGF group after the establishment of the model of AP. The other two groups of animals received the same volume of saline. At 6 h, 12 h and 24 h after induction of AP, 8 animals in each group were sacrificed respectively, 4 ml of blood sample was withdrawn from heart,2 ml for the analysis of amylase activity and 2 ml for MDA content in serum. Ascites was sucked with dry gauzes and was weighed thereafter. Changes of pancreas morphology were evaluated at every time point. The same part of pancreas was removed for measurement of MDA content, apoptotic index (AI) and histologic changes. Results Histologic injury of the animals in the APEGF group was milder than that in the AP group. Ascites weight in the AP-EGF group decreased significantly compared with that in the AP group at 12 h and 24 h 〔(4.53±1.29) g vs (6.58±1.47) g, (7.64±1.85) g vs (11.96±2.13) g,P<0.05,P<0.01〕. Amylase activity in the APEGF group also decreased significantly compared with that in the AP group at 12 h and 24 h 〔(142.0±8.3) U/L vs (187.9±10.4) U/L, (194.3±10.4) U/L vs (253.3±8.6) U/L, P<0.05,P<0.01〕. MDA content in plasm 〔(2.34±0.23) μmol/L vs (3.15±0.38) μmol/L, P<0.05〕 and in pancreas 〔(5.21±1.46) μmol/g vs (7.68±1.63) μmol/g, P<0.01〕 in the APEGF group decreased significantly compared with those in the AP group at 24 h. AI of pancreas in the APEGF group increased significantly compared withthatintheAPgroupafteroperation〔(16.22±3.53)%〖KG4vs (7.35±1.04)%, (11.67±2.40)% vs (4.81±0.86)%, (6.38±1.42)% vs (1.97±0.21)%, P<0.01〕. Conclusion EGF may accelerate the restoration of pathologic injury and alleviate the hemorrhage and edema of pancreas. It may also depress MDA content in plasm and in pancreas so that to lessen oxidative damage. EGF may protect pancreas by inducing cellular apoptosis.
Objective To explore the effects of recombinant human growth hormone (rhGH) on senile patients after pancreaticoduodenectomy. MethodsFortysix patients were divided into the therapeutic group (rhGH, n=17) and control group (n=29). Both were treated with parenteral nutrition. In the therapeutic group, rhGH (8 u/d) was given hypodermically for 7 days. After operation the levels of albumin, prealbumin, transferrin, and immunoglobulin were measured. Postoperative fatigue syndrome and the average length of stay in hospital were observed too. ResultsAfter operation the levels of albumin, prealbumin, transferrin, and immunoglobulin in the therapeutic group were significantly higher than those of control group. The degree of postoperative fatigue syndrome in the therapeutic group was less than that of control group. The average length of stay in hospital was significantly shortened. Conclusion The early application of rhGH in senile patients after pancreaticoduodenectomy can enhance immune function, reduce the incidence of infection, promote the postoperative recovery, shorten the average length of stay in hospital,decrease the mortality, increase the safety of operation and improve the postoperative life quality of senile patients.
【Abstract】ObjectiveTo discuss the technique for pancreas transplantation alone(PTA). MethodsEighty-eight SD rats were used as donors and recipients. The PTA was performed with enteric drainage(E-D group, n=22) or bladder drainage(B-D group, n=22). The donor’s abdominal aorta(splenic artery) and portal vein(splenic vein) were anastomozed with the recipient’s abdominal aorta(end-to-side) and left renal vein(end-to-end). Blood glucose, food intake, water intake and urine volume were recorded before transplantation and on the 1st day, 3rd day, 7th day, 14th day and 30th day after transplantation. Results The mean PTA time was (33.1±11.1) min (donors) and (51.7±14.7) min(recipients). The grafts experienced no warm ischemia and the mean cold ischemia time was (46.5±17.1) min. After the successful PTA, the recipients’ blood glucose decreased on the first day after transplantation and reached normal on the third day. Their food intake, water intake and urine volume decreased and became stable 14 days later. ConclusionSuccessful PTA can restore the pancreatic endocrine function in diabetic rats. It is very important to master the technique for the operation.
目的 探討經轉化生長因子-β1 ( TGF-β1) 基因修飾的未成熟樹突狀細胞(imDC) 預處理大鼠小腸移植受體后的外周血及移植腸浸潤T 細胞的變化及意義。方法 選用近交系F344/ N 和BN 大鼠建立全小腸異位移植模型,實驗分4 組(每組24 只) : 同基因移植組(BN-BN 組) 、異基因移植組( F344/ N-BN 組) 、異基因移植+ TGF-β1 基因轉染imDC 組( F344/ N-BN + TGF2β1 組) 和異基因移植+ TGF-β1 基因轉染imDC + FK506 組( F344/ N-BN + TGF-β1 + FK506 組) 。各組大鼠分別于術后3 、5 、7 d 各處死6 只,獲取大鼠靜脈血和移植腸。應用免疫組化SABC 法檢測受體鼠外周血及移植腸CD4 + 、CD8 + 、CD25 + 細胞和IL-4 的表達。同時行移植腸組織病理學檢查并觀察大鼠生存情況。結果 TGF-β1 修飾的DC 細胞能顯著抑制外周血及移植腸浸潤淋巴細胞CD4 + 、CD8 + 及CD25 + 的表達,并提高IL-4 的表達; 顯著延長受體大鼠的生存時間,但移植腸仍有排斥反應的病理組織學征象。結論 TGF-β1 修飾的DC 通過影響受體外周血及移植腸浸潤T 細胞對大鼠小腸移植發揮免疫抑制作用。
