Objective To observe and preliminarily explore the effects of Deferasirox (DFX) on lipid peroxidation and ferroptosis in human retinal endothelial cells (HREC). MethodsA cell experimental study. Divided the in vitro cultured HREC into normal glucose (NG) group, high glucose (HG) group, NG+DFX group, HG+DFX group, NG+DFX+ferric ammonium citrate (FAC) group, and HG+DFX+FAC group. Light microscope was used to observe the morphology of the cells; cell proliferation was detected by Cell Counting Kit-8 assay, and Calcein-AM staining was used to detect the unstable iron pool (LIP) content; enzyme-linked immunosorbent assay reader was used to detect the reactive oxygen species (ROS), malondialdehyde (MDA), glutathione (GSH), and oxidized glutathione (GSSG); Western blot was used to detect the relative protein expression of Glutathione Peroxidase 4 (GPX4) and Solute Carrier Family 7 Member 11 (SLC7A11). Two-tailed Student t test was used for comparison between the two groups; one-way ANOVA was used for comparison between multiple groups. ResultsCompared with the HG group and the HG+DFX+FAC group, the cell proliferation rate and the contents of GSH and the relative protein expression of GPX4, and SLC7A11 in the HG+DFX group were significantly increased, and the differences were statistically significant (F=150.70, 21.02, 26.09, 52.62; P<0.001). The contents of LIP, ROS, MDA, and GSSG were significantly decreased, and the differences were statistically significant (F=807.20, 16.94, 31.62, 19.21; P<0.001). ConclusionsHigh glucose significantly induces an increase in LIP, lipid peroxidation, and ferroptosis in HREC. Deferasirox inhibits lipid peroxidation and ferroptosis in HREC by downregulating LIP levels.
目的 探討18氟-脫氧葡萄糖(18F-FDG)雙探頭符合線路對非小細胞肺癌胸內淋巴結轉移的診斷價值,并分析其假陽性、假陰性診斷的原因。 方法 回顧性分析2010年12月-2012年6月非小細胞肺癌患者161例的臨床資料,其中腺癌122例,鱗癌23例,其他類型16例。分析患者術前18F-FDG雙探頭符合線路顯像對肺門、縱膈淋巴結的診斷結果(其中陽性53例,陰性108例),以術后病理診斷為金標準,評價其診斷價值。分析9例假陽性、26例假陰性診斷患者的吸煙史、慢性支氣管炎肺氣腫、肺結核病史、局限性肺炎、腫瘤標志物、淋巴結短徑、腫瘤原發灶T/N比值、外周血白細胞等指標。 結果 18F-FDG雙探頭符合線路單光子發射計算機斷層成像(SPECT)診斷非小細胞肺癌淋巴結轉移的靈敏度、特異性、準確性、陽性預測值、陰性預測值分別是62.9%、90.1%、78.3%、83%、75.9%。假陽性組患者慢支炎肺氣腫、局限性肺炎者高于真陽性組。假陰性組的淋巴結短徑、原發灶T/N比值小于真陽性組。 結論 18F-FDG雙探頭符合線路SPECT是術前診斷肺癌淋巴結轉移的有效手段;假陽性淋巴結與慢支炎肺氣腫、局限性肺炎有關;假陰性淋巴結與淋巴結短短徑小、原發腫瘤攝取18F-FDG低有關。
We assayed the levels of free radical and scavenger in the blood and lens of streptozotocin-in-duced diabetic SD rats, and found that the levels of lipoperoxide(LPO),MDA were higher than that of normal SD rats, and the total superoxygen dismutase (T-SOD), Cu-Zn-SOD) were lower that that of normal rats ( P lt;0.01 ). Simultaneous injection of streptozotocin and large dose of SOD could no avoid the occurence of diabetes mllitus, but did improve the metabolism of free radical in blood and lens. Hence, we think that large dose of SOD might be effective in preventing to development of diabetic cataract which is related to deterioration of free radical metabolism. This study aimed to assess the therapeutic effect of mannital administration on acute hemorrhagic necrotizing pancreatitis (AHNP), 28 Wistar rats were randomily divided into therapeutic group and control group after induction of AHNP by retrograde intraductal injection of 5 percent sodium taurocholate. The rats of therapeutic group received intravenous 20% mannital (1g/kg) through tail vein, once in 12 hours, until the end of experiment; control group received saline (5.0 ml/kg) with the same way. Blood of all the rats were collected from heart and the rats were killed after 96 hours. Results: lipid peroxide (LPO) in pancreatic tissue, LPO in serum, lactate dehydrogenase (LDH), alpha-1-antitrypsin (α1-AT), glutamicoxalacetic transaminase (GOT), necrotizing square of pancreatic tissue in the therapeutic group were significantly less than those in control group (P<0.05 or P<0.01). The damage to pancrease, heart, liver, kidney in the therapeutic group were lighter than those of the control group and the mortality was lower (P<0.05).Conclusions: Mannital can scavenge the oxygenderived free radicals and play a therapeutic role in AHNP.
摘要:目的: 評價機械通氣對胸腔內脈搏氧飽和度的影響。 方法 :以食道、氣管和降主動脈作為胸腔內脈搏氧飽和度的監測位點,將改制后的氧飽和度探頭分別固定于上述部位,并連接于同一監護儀上。純氧通氣,待上述氧飽和度容積波波形和讀數穩定,停止機械通氣30s。以錄像的方式記錄機械通氣停止前后30 s內食道、氣管和降主動脈SpO2容積波和讀數的變化。同時記錄舌SpO2。 結果 :機械通氣時,食道、氣管和降主動脈三個監測位點均可獲得異常高大的SpO2容積波;停止通氣時,異常高大的氧飽和度波形消失。食道、氣管和降主動脈脈搏容積波變異率分別為112%,74%,302%。降主動脈脈搏容積波的變異率明顯高于食道和氣管( 〖WTBX〗P <005)。機械通氣停止前后30s內食道、氣管和降主動脈的SpO2讀數變化無顯著差異(〖WTBX〗P >005)。 結論 :機械通氣對胸腔內食道、氣管和降主動脈氧飽和度讀數無影響,主要影響是脈搏容積波。且各位點間脈搏氧容積波受呼吸的影響不同。Abstract: Objective: To investigate the impact of mechanical ventilation on pulse oximetry in thoracic cavity. Methods : After dogs anesthesia induction and thoracotomy, pulse oximeters were simultaneously placed at esophagus, trachea, and descending aorta, and connected with the same monitor for SpO2 monitoring. During ventilation with 100% oxygen, the mechanical ventilator was temporarily switched off for 30 seconds after high quality PPG waveforms and SpO2 readings were obtained. SpO2 signals and readings from esophagus (SeO2), trachea (StraO2), descending aorta (SDAO2) shown on the monitoring screen were recorded by the SONY video before and after stopventilation. And StonO2 were also recorded. Results : Abnormally largeamplitude PPG waves were found in normal waves at monitoring sites of esophagus, trachea, and descending aorta in all animals during ventilation; however, they disappeared without ventilator. The variation rate in ventilationinduced PPG amplitude were 112%, 74%, 302% at esophagus, trachea and descending aorta respectively. The PPG amplitude variation rate from SDAO2 was higher than that from SeO2 and StraO2 (〖WTBX〗P <005). However, the SpO2 readings obtained from pulse oximetries in all sites were no significantly statistical difference within 30s before and after temporarily stopventilation (〖WTBX〗P >005). Conclusion : Abnormally amplitude PPG waveforms from oximetry probe placed at esophagus, trachea, and descending aorta were induced by ventilation. The Variation rate in ventilationinduced PPG amplitude was various at different monitoring sites. The SpO2 readings from esophagus, trachea, and descending aorta were not significantly contaminated by ventilation.
Objective To observe the influence of the expression of CD18 on the neutrophile and the leukocyte adhesion to retinal vascular endothelium by hypoxia-inducible factor-1 alpha (HIF-1alpha;) in early diabetic retinopathy rats. Methods Male Sprague-Dawley rats received intraperitoneal injection of streptozotocin to induce diabetes model. 18 diabetic rats were divided into 3 groups randomly after 2 months of diabetes induction, including diabetic group (group B), HIF-1alpha; anti-sense oligonucleotides (ASODN) injection group (group C) and HIF-1alpha; sense oligonucleotides (SODN) injection group (group D), the age and weigh matched health rats were chosen as control group (group A), with 6 rats in each group. Then group A and B rats received 5% glucose solution caudalis veins injection, group C and group D rats received HIF-1alpha; ASODN and HIF-1alpha; SODN caudalis veins injection, respectively(025 mg/kg).The level of CD18 on the neutrophil isolated from the peripheral blood was measured by flow cytometry. Retinal leukostasis was quantified with acridine orange leukocyte fluorography. Results The percentage of CD18 positive neutrophil cell was(44.93plusmn;3.60)% in group B,(18.66plusmn;1.52)% in group A,(31.66plusmn;4.72)% in group C,(51.00plusmn;5.66)% in group D. Compared with each other groups,the differences are statistically significant (F=42.46, Plt;0.001). The number of positive staining cells of retinal leukocyte was (46.16plusmn;10.68)in group A,(133.83plusmn;20.43)in group B,(99.83plusmn;9.28)in group C,(121.33plusmn;10.23) in group C. Compared group B with group C,the number of positive staining cells raised about 2.89 times;compared group B with group C and D,the differences are statistically significant (P=0.12,95% confidence interval -3.69~28.69). Conclusions In vivo, HIF-1alpha; can decreased the expression of CD18 on neutrophils from diabetic ratsprime; peripheral blood and the collection of retinal leukostasis in the diabetic animals. HIF-1alpha; may serve as a therapeutic target for the treatment and/or prevention of early diabetic retinopathy. (Chin J Ocul Fundus Dis,2008,24:268-271)
【摘要】 目的 通過總結耐甲氧西林金黃色葡萄球菌(meticillin-resistant staphylococcus aureus, MRSA)感染皮膚疾病患者的護理經驗總結,探尋控制皮膚疾病感染的有效措施。 方法 對2009年1-10月收治的30例MRSA感染皮膚疾病患者的護理資料進行回顧分析。 結果 通過精心治療、護理,30例患者皮損均治愈。 結論 采取嚴格的接觸隔離措施,加強手衛生,對癥選擇敷料及換藥,正確處理醫用垃圾,加強與患者或家屬的溝通交流,認真進行衛生宣教等,能有效的控制MRSA感染,提高臨床痊愈率。【Abstract】 Objective To investigate the appropriate nursing for patients with skin disease due to methicillin-resistant staphylococcus aureus (MRSA) infection. Methods Careful nursing had been taken to 30 patients with skin disease due to MRSA infection from January to October in 2009. Results Skin lesions of 30 patients were cured through conscientious treatment and intensive nursing. Conclusion Strict contact isolation measures, strengthened hand hygiene, careful dressing, correct medical waste disposal, communication with patients or their families, and health education can effectively control the MRSA infection and improve the clinical cure rate.
