目的　采用液相色譜-串聯質譜法測定人血漿中二甲雙胍的濃度。方法　血漿樣品用乙腈（含0.1%甲酸）沉淀蛋白后用二氯甲烷反洗后進行分析。使用Agilent C8（75 mm×4.6 mm，3.5 μm）色譜柱。流動相：A泵：5 mmol/L醋酸銨（三乙胺調pH值至7.5），B泵：乙腈。線性梯度洗脫，流速0.4 mL/min。采用電噴霧離子源，多反應離子監測。用于定量分析的離子對二甲雙胍為130.2/71.1，內標嗎啉胍為172.2/60.2。結果　線性范圍為50～2 000 ng/mL，最低定量限為50 ng/mL，預處理回收率為81.7%～98.0%，二甲雙胍的基質效應＜9.97%，日內和日間相對標準偏差均＜5.2%。結論　液相色譜-串聯質譜法快速、簡便、靈敏度高，是一種適用于人血漿中藥物濃度的測定及藥物動力學和生物利用度研究的方法。
